    Microbial Community Engineering for Clean Water, Bioenergy Generation, and Resource Recovery

    Abstract
    Microbial bioprocesses are a cornerstone of our urban water quality control infrastructure. They are also increasingly viewed as a crucial platform in the emerging circular bioeconomy to recover valuable resources (e.g. clean water, energy, nutrients, biomaterials, bio-based chemicals) from organic carbon and nutrient rich societal waste streams. Despite their environmental and economic importance, surprisingly little is known about the nature or “design rules” of the complex microbial communities that underlie emergent function in these bioprocesses. In this talk, I will detail of our efforts to deepen understanding of diversity, metabolic versatility, community assembly mechanisms, and ecological selection pressures in the microbial world in order to inspire novel resource efficient environmental bioprocesses, with a particular focus on nitrogen (N) and phosphorus (P) removal and recovery. Our efforts in this area address the grand challenge of preventing nutrient pollution responsible for a vast array of environmental and public health problems, including eutrophication of nutrient limited water bodies, greenhouse gas (nitrous oxide) production, and direct adverse effects to human health (e.g. methemoglobinemia caused by nitrate.). First, I will present evidence that newly discovered complete ammonia oxidizing bacteria (comammox) are a crucial but previously unrecognized biocatalyst for high rate and energy efficient nitrification at water resource recovery facilities. However, our work also shows that comammox may impede ecological selection for anaerobic ammonia oxidation (anammox) in mainstream shortcut N removal bioprocesses via competition for metabolic intermediates. I will then introduce an emerging paradigm in which societal waste streams are viewed not as costly energy sinks but as feedstocks for energy or resource recovery. I will present efforts in line with this paradigm towards development of a novel mixed-culture bioprocess for microbial production and capture of nitrous oxide as a source of bioenergy, with concurrent recovery of P as a fertilizer. Genome-resolved ‘omics’ analyses of the underlying microbiome in this bioprocess have revealed a surprisingly high prevalence of incomplete denitrification pathways, with far reaching implications for understanding microbial nitrous oxide production and metabolic division of labor in both engineered and natural systems. Finally, I will conclude by highlighting our efforts to elucidate the ecology and genomic context of emerging “genetic contaminants” in the form of antibiotic resistance genes in engineered bioprocesses. Taken together, these results demonstrate the utility of coupling fundamental microbial ecology research to novel bioprocess development.

    Biography
    George Wells is an Assistant Professor in the Department of Civil and Environmental Engineering at Northwestern University, where he directs the Environmental Biotechnology and Microbial Ecology Laboratory. His primary research interests are resource and energy recovery from wastewater and other urban “waste” streams, microbial nitrogen and phosphorus cycling and shortcut biological nutrient removal processes, microbial ecology of engineered and impacted natural systems, biofilm structure-function relationships, and microbial greenhouse gas production. George received his B.S. in Chemical Engineering and B.A. in Environmental Engineering from Rice University in 2004. After a short period working at BP Chemicals in Naperville, Illinois, George joined the Department of Civil and Environmental Engineering at Stanford University, where he completed his MS (2006) and PhD (2011) under Dr. Craig Criddle and Dr. Chris Francis. Prior to joining Northwestern University in the fall of 2013, George spent nearly 2.5 years as a postdoctoral scholar under Dr. Eberhard Morgenroth at Eawag- the Swiss Federal Institute of Aquatic Science and Technology (near Zürich, Switzerland).
