Thursday, March 10th4:00-5:00PM, Tech L361Title:"Micro and Nano-Scale Biomaterials for Stem Cell-Based Tissue Regeneration and Cancer Treatment"Abstract:Stem cells have been proposed as promising cell-based medicine for facilitating tissue regeneration and cancer treatment. In the past years, the practice of stem cell culture and differentiation to date has been done relatively low efficiency (hanging drop method) or not physiologically relevant for embryonic stem cells (2D, 3D bulk gel). In this study, we developed a novel high throughput coaxial electrospray technology to generate microcapsules with a hydrogel shell of alginate and an aqueous liquid core of cells. We proved that murine embryonic stem (ES) cells can be encapsulated in the liquid core with high viability and proliferate to form single aggregate in each microcapsule with improved pluripotency.In terms of implantation of therapeutic cells, it is difficult to achieve minimally invasive injectable cell delivery while maintaining a high allogeneic cell survival and function in vivo. Here we reported a novel conformal nanoscale biomaterial technology to encapsulate and deliver cells. This nanoscale biomaterial encapsulation is based on electrostatic deposition of positively and negatively charged biomaterials and it gives a semi-permeable physical shield on the surface of cells, which will not prevent nutrients and oxygen transported to cells, but could possibly minimize the cell-cell contact that activates immune cell like macrophages, which will reduce undesirable immune response and inflammation.
